اثر هم‌افزایی تمرین هوازی و ویتامین D₃ بر sFasL ،sFas و نسبت آن‌ها در بافت کلیه رت‌های نر دیابتی

چکیده

زمینه و هدف: دیابت نوع ۲ (T2D) یک اختلال متابولیکی است که می‌تواند عملکرد کلیه را تحت تأثیر قرار دهد. هدف این مطالعه بررسی اثرات تعاملی تمرین هوازی تداومی با شدت متوسط (MICT) و ویتامین D₃ بر عملکرد کلیه در رت‌های مبتلا به T2D بود. روش پژوهش: چهل سر رت نر ویستار (۸ هفته، وزن ۲۵۰–۳۰۰ گرم) پس از دوره سازگاری، به گروه‌های رژیم غذایی معمولی (n=8) و پرچرب (n=32) تقسیم شدند. T2D با ۶ هفته رژیم پرچرب و یک نوبت تزریق استرپتوزوتوسین القاء شد. سپس رت‌ها به پنج گروه هشت‌تایی تقسیم شدند که شامل کنترل سالم، کنترل دیابتی، دیابت با تمرین هوازی، دیابت با تزریق ویتامین D₃ و دیابت با تمرین هوازی و تزریق ویتامین D₃ بودند. MICT به مدت ۸ هفته (دویدن ۳۰–۶۰ دقیقه، با شدت ۵۵–۶۵٪ حداکثر سرعت دویدن) و ویتامین D₃ از طریق تزریق زیرجلدی، با دوز ۱۰,۰۰۰ IU/kg، یک‌بار در هفته تجویز شد. در پایان، سطوح سرمی گلوکز، انسولین و پروتئین‌های کلیوی sFas و sFasL اندازه‌گیری شدند. یافته‌ها: در گروه کنترل دیابتی، در مقایسه با گروه کنترل سالم، سطوح sFas و نسبت sFas/sFasL به‌طور معنی‌داری بالاتر و سطوح sFasL پایین‌تر بود (P < 0.001)، که بیانگر فعال‌سازی بیش‌ازحد آپوپتوز است. همچنین، پس از 8 هفته MICT، در گروه‌های تمرینی نسبت به گروه کنترل دیابتی، سطوح sFas و نسبت sFas/sFasL به‌طور معنی‌داری کاهش و سطوح sFasL افزایش یافت. بیشترین کاهش آپوپتوز در گروه تعاملی مشاهده شد (P < 0.001). علاوه بر این، این مداخلات موجب تعدیل سطوح سرمی انسولین، گلوکز و شاخص HOMA-IR گردید. نتیجه‌گیری: T2D موجب افزایش فرآیند آپوپتوز در بافت کلیه می‌شود. به‌کارگیری MICT همراه با ویتامین D₃ باعث بهبود عملکرد کلیه و کاهش آپوپتوز در رت‌های دیابتی گردید. این نتایج نشان می‌دهد که ادغام این دو مداخله می‌تواند راهبردی مؤثر برای کاهش عوارض کلیوی ناشی از دیابت فراهم آورد.

Abstract

Background & Purpose: Type 2 diabetes (T2D) is a metabolic disorder that can impair kidney function. This study aimed to investigate the interactive effects of moderate-intensity continuous aerobic training (MICT) and vitamin D₃ on Kidney function in T2D rats. Methods: Forty male Wistar rats (8 weeks old, 250–300 g) after an acclimatization, were divided into a standard diet group (n=8) and a high-fat diet group (n=32). T2D was induced in the high-fat diet group by 6 weeks of high-fat feeding followed by a single injection of streptozotocin. Subsequently, rats were divided into five groups of eight: Healthy Control, Diabetic Control, Diabetic with Aerobic Training, Diabetic with Vitamin D₃ injection, and Diabetic with combined Aerobic Training and Vitamin D₃. The MICT protocol consisted of treadmill running for 30–60 minutes per session at 55–65% of maximal running speed for 8 weeks, and Vitamin D₃ was administered subcutaneously at a dose of 10,000 IU/kg once weekly. At the end of the intervention, serum glucose, insulin, and Kidney proteins sFas and sFasL were measured. Results: In the Diabetic Control group, compared to the Healthy Control group, serum sFas and the sFas/sFasL ratio were significantly higher, while sFasL were lower (P < 0.001), indicating excessive apoptosis activation. Furthermore, following 8 weeks of MICT, the training groups exhibited significantly reduced sFas and sFas/sFasL ratio, and increased sFasL compared to the Diabetic Control group. The greatest reduction in apoptosis was observed in the combined intervention group (P < 0.001). Additionally, these interventions effectively modulated serum insulin, glucose levels, and the HOMA-IR index. Conclusions: T2D promotes apoptosis in Kidney tissue. The combination of MICT and vitamin D₃ improved Kidney function and reduced apoptosis in diabetic rats. These findings suggest that integrating these two interventions may represent an effective strategy to mitigate diabetes-induced Kidney complications.
