تغییرات بیان ژن‌های UCP1 و آیریزین بافت چربی موش‌های صحرایی نر چاق متعاقب تمرین استقامتی و تحریک الکتریکی با محدودیت کالری

چکیده

هدف: چاقی عامل خطر بسیاری از بیماری‌های‌ قلبی‌عروقی، پرفشار خونی، افزایش چربی، دیابت و سرطان است و ژن‌های مهمی مانند UCP1 و آیریزین در آن نقش دارند. لذا هدف از پژوهش حاضر بررسی تغییرات بیان ژن‌های UCP1 و آیریزین بافت چربی موش‌های صحرایی نر چاق متعاقب تمرین استقامتی و تحریک الکتریکی با محدودیت کالری بود. روش‌ پژوهش: در این مطالعۀ تجربی کنترل‌ شده با گروه شاهد 35 سر موش صحرایی نر ویستار (8 هفته‌ای با وزن 19±200 گرم) پس از القا چاقی و افزایش وزن به‌طور تصادفی به 5 گروه 7 تایی کنترل، چاق روزه‌دار، روزه‌دار-تمرین استقامتی، روزه‌دار-تحریک الکتریکی و روزه‌دار-تمرین استقامتی-تحریک الکتریکی تقسیم شدند. گروه‌های مداخله برای یک دوره 4 هفته‌ای تحت فعالیت ورزشی استقامتی (با سرعت 10 تا 20 متر/دقیقه و مدت 20 تا 40 دقیقه)، تحریک الکتریکی (دستگاه فوت شوک برای 5/0 میلی‌آمپر و 20دقیقه) و روزه‌داری (8 به 16 ساعت) قرار گرفتند. پس از تمرین و بی‌هوشی، نمونه‌بردای بافت چربی صورت گرفت و پس از انجام فرایندهای ملکولی بیان ژن‌ها با استفاده از دستگاه Real time-PCR اندازه‌گیری شد. برای آنالیز داده‌ها از آزمون تحلیل واریانس دو‌طرفه در سطح معنی‌داری 05/0>p و نرم‌افزار گراف‌پد استفاده شد. یافته‌ها: نتایج نشان داد تمرین استقامتی منجر به افزایش معنی‌دار (0007/0=P) بیان ژن‌های UCP1 و افزایش غیرمعنی‌دار آیریزین در موش‌های صحرایی چاق روزه‌دار نسبت به گروه چاق روزه‌دار شد (766/0=P ). همچنین تحریک الکتریکی در هر دو متغیر کاهش معنی‌داری نسبت به گروه کنترل نشان داد (0001/0=P). از طرفی تحریک الکتریکی و تلفیق آن با تمرین استقامتی منجر به افزایش معنی‌دار بیان ژن آیریزین نسبت به کلیه گروه‌ها شد (0001/0=P) اما در ژنUCP1  کاهش معنی‌داری نسبت به گروه کنترل و چاق روزه‌دار نشان داد (0001/0=P). نتیجه‌گیری: به‌نظر می‌رسد تمرین استقامتی در حین روزه‌داری با افزایش مقادیر ژن‌های پروتئین UCP1 و آیریزین در نمونه‌های چاق در تبدیل بافت چربی سفید به قهوه‌ای و کاهش اثرات منفی آن می‌تواند موثر باشد.

چکیده [English]

Aim:      Obesity is a risk factor for many cardiovascular diseases, hypertension, hyperlipidemia, diabetes, and cancer, and genes such as UCP1 and irisin play a significant role in it. Therefore, the aim of this study was to investigate the changes in the expression of UCP1 and irisin genes in the adipose tissue of obese rats following endurance exercise and electrical stimulation with caloric restriction. Methods: In this controlled experimental study with a control group, 35 male Wistar rats after induced obesity (8 weeks old, weighing 200±19 g) were randomly divided into 5 groups of 7: control, obese-fasted, fasted-endurance exercise, fasted-electrical stimulation, and fasted-endurance exercise-electrical stimulation. The intervention groups underwent endurance exercise (10-20 m/min for 20-40 minutes), electrical stimulation (foot shock device for 0.5 mA and 20 minutes), and fasting (8-16 hours) for a period of 4 weeks. After exercise and anesthesia, adipose tissue samples were taken and gene expression was measured using Real-time PCR. Data were analyzed using two-way ANOVA with a significance level of p<0.05 and GraphPad software. Results: The results showed that endurance exercise significantly increased (P=0.0007) the expression of UCP1 gene and non-significantly increased irisin in obese fasted rats compared to the obese fasted group (p=0.766). Electrical stimulation also showed a significant decrease in both variables compared to the control group (P=0.0001). On the other hand, electrical stimulation and its combination with endurance training led to a significant increase in irisin gene expression compared to all groups (P=0.0001), but in the UCP1 gene, it showed a significant decrease compared to the control and fasting obese groups (P=0.0001). Conclusion: It seems that endurance exercise during fasting can be effective in converting white adipose tissue to brown adipose tissue and reducing its negative effects by increasing the levels of UCP1 and irisin proteins in obese samples.
