Background& purpose: It has been shown that regular exercise is effective in maintaining and improving heart function. Therefore, this study investigated the effect of running training supplemented with the hydro-alcoholic extract of Garhaqat fruit on glycemic indices and the expression of miR-195 in the heart tissue of rats. Methods: 24 male Wistar rats (weight 20±200 gr and age of 8 weeks) were randomly divided into four groups: control, aerobic training, supplementation, and training+supplementation groups. The rats in the supplemented groups received 250 mg of Qaraqat fruit extract per kilogram of body weight dissolved in 0.2 cc of normal saline in the form of oral gavage. The training protocol was obtained on the treadmill five days per week with the principle of overload in the first week at a speed of 5-10 m/min for 10-15 minutes and in the eighth week at a speed of 18-24 m/min for 60 minutes. Glucose and insulin, insulin resistance, and miR-195 expression of cardiac tissue were measured. One-way ANOVA and Bonferroni post hoc test were used at a significant level of P ≥0.05. Results: A significant increase in the expression of miR-195 in the aerobic exercise group compared to the control groups (P<0.001) and supplement (P=0.010), and the expression of miR-195 in the exercise+supplement group compared to the control group ( P=0.002) was observed. However, no significant difference was observed in glucose and insulin indices and insulin resistance (P<0.05). Conclusions: The results of the present study showed that aerobic training leads to a significant increase in miR-195 gene expression, but this increase is less with Karaqat supplementation than with aerobic exercise. In addition, the combination of the supplementation of Karaqat with aerobic exercise may reduce some of the increase in miR-195 and thereby reduce oxidative stress induced by aerobic exercise.
