اثرات 8 هفته تمرین هوازی تداومی و تناوبی به همراه مصرف مکمل رزوراترول بر بیان ژن‌هایBeclin-1 و LC3I-II بافت قلب رت‌های نر دیابتی

چکیده

هدف: کاردیومیوپاتی دیابتی عارضه‌ای از دیابت نوع دو است که می‌تواند منجر به اتوفاژی سلولی عضله قلب از طریق پروتئین‌های Beclin-1 و LC3 شود. هدف از این پژوهش بررسی اثرات 8 هفته تمرین تداومی و تناوبی هوازی به همراه مصرف مکمل رزوراترول بر بیان ژن‌هایBeclin-1  و LC3 بافت قلب رت‌های نر دیابتی بود. روش شناسی: به این منظور تعداد 56 سر رت نر نژاد ویستار با میانگین (سن50-40 هفته و وزن430-373 گرم) به طور تصادفی به 8 گروه (7n=) کنترل (C)، دیابتی(D)، سالین(S)، رزواترول(Sup)، تمرین‌تداومی(E)، تمرین‌تناوبی HIIT))، تمرین تداومی/مکمل (E+Sup)، تمرین تناوبی/ رزوراترول (HIIT+Sup) تقسیم شدند. دیابت با تزریق درون صفاقی استرپتوزوتوسین با دوز 50 میلی‌گرم به ازای هر کیلوگرم وزن بدن انجام شد. رت‌های تمرین تداومی سه روز در هفته به مدت 8 هفته و با سرعت 15متر بر دقیقه تمرین کردند. پروتکل تمرین تناوبی نیز شامل 10 ست یک دقیقه‌ای با شدت 50 درصد و دو دقیقه استراحت بین ست‌ها بود و با سرعت 14 متر در دقیقه شروع و هر هفته دو متر در دقیقه به سرعت افزوده ‌شد. داده‌ها با استفاده از تحلیل واریانس دو راهه در سطح 05/0≥ α تحلیل شدند. یافته‌ها: نتایج نشان داد Beclin-1  و LC3 در گروه القای دیابتی کاهش معنی‌داری نسبت به گروه (C) سالم داشت (001/0P=). در مقایسه با گروه دیابتی، گروه دیابتی+تمرین+مکمل افزایش معنی‌داری را در Beclin-1 و  LC3نشان داد (001/0 P=). نتیجه‌گیری: تمرینات ورزشی به همراه مصرف مکمل رزوراترول می‌تواند باعث افزایش فاکتورهای اتوفاژی Beclin-1 و LC3 شود که موجب مهار آپوپتوز و کاهش آسیب بافت قلب در بیماران دیابتی می‌شود.

Aim: Diabetic cardiomyopathy is a complication of type 2 diabetes that can lead to myocardial cell autophagy through Beclin-1 and light chain 3 proteins. The aim of this study was to evaluate the effects of 8 weeks of continuous and intermittent aerobic exercise with resveratrol supplementation on Beclin-1 and LC3 gene expression in the heart tissue of diabetic male rats. Methods:  For this purpose, 56 male Wistar rats with mean (age 40-50 weeks and weight 430-373 g) were randomly divided into 8 groups (n = 7) control (C), diabetic (D), saline (S), Resveratrol (Sup), Continuous exercise (E), Interval exercise (HIIT), Continuous exercise/Supplement (E+Sup), Intervalt exercise/Resveratrol (HIIT+Sup).  Diabetes was diagnosed by intraperitoneal injection of streptozotocin at a dose of 50 mg / kg body weight. Continuous training rats trained three days a week for 8 weeks at a speed of 15 meters per minute.  The intermittent training protocol also included 10 one-minute sets with an intensity of 50% vo2max on the treadmill and two minutes of rest between sets, starting at 14 meters per minute and increasing rapidly by two meters per minute each week.  Data were analyzed using two-way analysis of variance at the α (α ≥0.05) level. Results:  The results showed that Beclin-1 and LC3 in the diabetic induction group had a significant decrease compared to the healthy control group (P = 0.001). Compared with the diabetic group, the diabetic + exercise + supplement group showed a significant increase in Bcline-1 and LC3 (P = 0.001). Conclusion:   Exercise combined with resveratrol supplementation can increase Beclin-1 and LC3 autophagy factors” which inhibit apoptosis and reduce heart tissue damage in diabetics.
